A 65 year-old wheelchair bound man died on Sunday morning when the farmhouse at Goedemoetsfontein in the Kragga Kamma Area outside Port Elizabeth burnt to the ground.
Police warrant officer Alwan Labans says another family member who was also in his 60s made it out alive.
Labans says the building was completely destroyed and that forensics experts are trying to determine the cause of the blaze.
He confirmed to AlgoaFM news that they'd also combed the ruins after 58 year-old Margaret Bentley was reported missing.
He said there were definitely no remains of any other person in the rubble.
Family members are concerned for the welfare of Bentley and she's asked to make contact with her daughter Monique - or her nearest police station.
