Five people, including a four-month-old baby, were killed in a head-on collision on the N2 between Nananga and Makhanda on Sunday.
The Eastern Cape Transport Department said six other people sustained serious injuries following the collision between two cars.
Spokesperson, Makhaya Komisa, said emergency service has treated the injured passengers and moved them to nearby hospitals
He said the Transport and Community Safety MEC Xolile Nqatha has expressed his heartfelt condolences to the grieving families and wished the injured passengers a speedy recovery.
"MEC Nqatha continues to call on motorists to adhere to the rules of the road as most accidents that have occurred in the province have been caused by human behaviour," Komisa said.
