Spokesperson, Colonel Sibongile Soci says the suspect was arrested in Dutywa on Friday.
She says the man was driving a Toyota Quantum with a trailer when patrolling police members became suspicious and stopped the vehicle.
Upon searching the trailer, police found 38 bags full of dagga weighing 745kg.
Soci says the suspect was en route from Mount Frere to Port Elizabeth.
The man will appear in the Dutywa Magistrate's Court on Monday.
