Two people lost their lives on the R-390 between Hofmeyer and Cradock in the Eastern Cape.
Transport Department spokesperson, Unathi Binqose, said the driver apparently lost control of the vehicle which overturned on Friday morning.
He said the female driver and a male passenger were killed while two other people sustained light injuries.
Binqose said it was alleged that speeding and fatigue may have contributed to the crash.
Police have opened a culpable homicide docket.
