Two people were killed after two trucks and a taxi collided at the N2 intersection near Deal Party.
A spokesperson for Provincial Traffic Unathi Binqose says the accident happened shortly after midnight at the M4 intersection in Gqeberha.
He says two trucks were travelling in the same direction when one allegedly rear-ended the other.
Binqose says as a result of the impact a trailer disconnected from one of the trucks and obstructed the lane to oncoming traffic.
A minibus taxi with three occupants then hit the trailer head-on.
Binqose says the driver, who hails from the Western Cape was trapped in the wreckage and declared dead on the scene.
One of his passengers died on his arrival at the hospital while another sustained critical injuries.
One of the truck drivers was also seriously injured.
A case of culpable homicide is under investigation.
