Three rhinos were shot at the Sibuya Game Reserve near Kenton-on-Sea in the Eastern Cape in the early hours of Thursday morning.
Marketing manager, Charlene Watson, said two rhinos were killed and one was dehorned.
A third rhino was still alive but the animal's horn had also been removed.
Watson said police were at the reserve and a helicopter had been brought in to assist with the investigation.
Spokesperson, Lieutenant Luvuyo Mjekula, said police were also searching for suspects in and around the game reserve.
Mjekula said the police unit responsible for rhino poaching and the K9 unit were also on scene
"Police are still at the scene as we speak and other authorities, the dog unit is there, they are conducting a search as we speak for the suspects. There is suspicion they might still be in the vicinity but no one has been arrested at this moment but police are still searching"
Anyone who may have information on the poaching incident has been asked to contact the Port Alfred police.
Meanwhile, this is second major poach incident in the province in recent weeks.
The owner of the game reserve outside Cradock offered a substantial reward after two white rhinos were poached and killed on the reserve about a week ago.
A four- year-old bull calf and it's mother, who was pregnant, were found dead on the Tam Safaris Game Reserve last week Friday with their horns sawn off.
They had both been darted and tranquilized.
