Six people – including the two bus drivers – have died when the vehicles crashed on the N2 near Mossel Bay in the early hours of Saturday morning.
Police spokesperson Sergeant Chris Spies said 25 others were injured when the buses, a Williams Coach and an Intercape bus, collided at around 1.30 in the morning.
The Mossel Bay municipality said a section of the N2 would remain closed as well as both lanes in the direction of George to Mossel Bay between the Langeberg Mall and Dana Bay.
Both lanes of the N2 direction Mossel Bay to George were open.
On Saturday morning, the municipality said an investigation team was on route from Cape Town to document and investigate the accident.
